Biomedical Optics Laser speckle rheology (LSR) offers a new noncontact optical approach. LSR uses an inexpensive laser—similar to a common laser pointer to shine laser light on tissue—plus a camera to image speckle patterns reflected from light-scattering particles in the sample. When laser light which has been scattered off a rough surface falls on another surface, it forms an "objective speckle pattern". Laser Speckle Rheology (LSR) is an optical technique to evaluate the viscoelastic properties by analyzing the temporal fluctuations of backscattered speckle patterns. Variations of optical absorption and reduced scattering coefficients further modulate speckle fluctuations, posing a critical challenge for quantitative evaluation of viscoelasticity. We introduce a novel technology, termed laser speckle micro-rheology (LSM) that offers the unique capability to map the viscoelastic properties of the tissue microenvironment. This innovation will provide important insight into mechano-biology of disease progression. Laser speckle microrheology is used to determine a mechanical property of a biological tissue, namely, an elastic modulus. Speckle frames may be acquired by illuminating a coherent light and effectively measure tissue stiffness.
